    Methyl Coenzyme M Reductase (mcrA) Gene Abundance Correlates with Activity Measurements of Methanogenic H2/CO2-Enriched Anaerobic Biomass

    Biologically produced methane (CH4) from anaerobic digesters is a renewable alternative to fossil fuels, but digester failure can be a serious problem. Monitoring the microbial community within the digester could provide valuable information about process stability because this technology is dependent upon the metabolic processes of microorganisms. A healthy methanogenic community is critical for digester function and CH4 production. Methanogens can be surveyed and monitored using genes and transcripts of mcrA, which encodes the α subunit of methyl coenzyme M reductase – the enzyme that catalyses the final step in methanogenesis. Using clone libraries and quantitative polymerase chain reaction, we compared the diversity and abundance of mcrA genes and transcripts in four different methanogenic hydrogen/CO2 enrichment cultures to function, as measured by specific methanogenic activity (SMA) assays using H2/CO2. The mcrA gene copy number significantly correlated with CH4 production rates using H2/CO2, while correlations between mcrA transcript number and SMA were not significant. The DNA and cDNA clone libraries from all enrichments were distinctive but community diversity also did not correlate with SMA. Although hydrogenotrophic methanogens dominated these enrichments, the results indicate that this methodology should be applicable to monitoring other methanogenic communities in anaerobic digesters. Ultimately, this could lead to the engineering of digester microbial communities to produce more CH4 for use as renewable fuel

    #RetroPIDs: the missing link to the foundation of biodiversity knowledge

    The Biodiversity Heritage Library (BHL) will soon upload its 60 millionth page of open access biodiversity literature onto the BHL website and the BHL's Internet Archive Collection. The BHL’s massive repository of free knowledge includes content that is available nowhere else online, as well as accessible versions of content that are locked behind paywalls elsewhere. If we are to continue to expand our understanding of life on Earth, we must ensure that the foundation of biodiversity knowledge provided by BHL is discoverable by the tools we rely on to navigate the ever-expanding internet. These tools – search engines and their algorithms – preferentially deliver (and rank) content with good metadata and persistent identifiers (PIDs). In modern online publishing, PID assignment and linking happens at the point of publication: DOIs (Digital Object Identifiers) for publications, ORCIDs (Open Researcher and Contributor IDs) for people, and RORs (Research Organization Registry IDs) for organisations. The DOI system provided by Crossref (the DOI registration agency for scholarly content) delivers reciprocal citations, enabling convenient clicking from article to article, and citation tracking, enabling authors and institutions to track the impact and reach of their research output. Publications that lack PIDs, which include the vast majority of legacy literature, are hard to find and sit outside the linked network of scholarly research. This makes it nearly impossible to determine whether they are being cited, let alone viewed, mentioned, shared or liked. At TDWG 2020, Page 2020, Kearney 2020, Richard 2020 (and 2019, Page 2019b, Page 2019a, Kearney 2019b, Kearney 2019a and 2018, Kearney 2018), we emphasised the need to bring the historic biodiversity literature into the modern linked network of scholarly research. In October 2020, BHL launched a new working group to do exactly this. The BHL Persistent Identifier Working Group (Team #RetroPID) brings together expertise from across BHL’s global community. Over the past year, we have worked tirelessly to make it easier to find, cite, link, share and track the content on BHL, adding article-level metadata to journals and retrospectively assigning DOIs (#RetroPIDs). Most importantly, we have developed the tools and documentation that will enable the entire BHL community to take contributed content from “just” accessible to persistently discoverable. This paper will detail our efforts to retrofit the historic literature (a square peg) into the modern PID system (a round hole) and will present both the achievements and the challenges of this important work

    State-Dependent Effects of Ventromedial Prefrontal Cortex Continuous Thetaburst Stimulation on Cocaine Cue Reactivity in Chronic Cocaine Users

    Cue-induced craving is a significant barrier to obtaining abstinence from cocaine. Neuroimaging research has shown that cocaine cue exposure evokes elevated activity in a network of frontal-striatal brain regions involved in drug craving and drug seeking. Prior research from our laboratory has demonstrated that when targeted at the medial prefrontal cortex (mPFC), continuous theta burst stimulation (cTBS), an inhibitory form of non-invasive brain stimulation, can decrease drug cue-related activity in the striatum in cocaine users and alcohol users. However, it is known that there are individual differences in response to repetitive transcranial magnetic stimulation (rTMS), with some individuals being responders and others non-responders. There is some evidence that state-dependent effects influence response to rTMS, with baseline neural state predicting rTMS treatment outcomes. In this single-blind, active sham-controlled crossover study, we assess the striatum as a biomarker of treatment response by determining if baseline drug cue reactivity in the striatum influences striatal response to mPFC cTBS. The brain response to cocaine cues was measured in 19 cocaine-dependent individuals immediately before and after real and sham cTBS (110% resting motor threshold, 3600 total pulses). Group independent component analysis (ICA) revealed a prominent striatum network comprised of bilateral caudate, putamen, and nucleus accumbens, which was modulated by the cocaine cue reactivity task. Baseline drug cue reactivity in this striatal network was inversely related to change in striatum reactivity after real (vs. sham) cTBS treatment (ρ = -.79; p < .001; R2Adj = .58). Specifically, individuals with a high striatal response to cocaine cues at baseline had significantly attenuated striatal activity after real but not sham cTBS (t9 = -3.76; p ≀ .005). These data demonstrate that the effects of mPFC cTBS on the neural circuitry of craving are not uniform and may depend on an individual’s baseline frontal-striatal reactivity to cues. This underscores the importance of assessing individual variability as we develop brain stimulation treatments for addiction

    Exploring patient information needs in type 2 diabetes: A cross sectional study of questions

    This study set out to analyze questions about type 2 diabetes mellitus (T2DM) from patients and the public. The aim was to better understand people's information needs by starting with what they do not know, discovered through their own questions, rather than starting with what we know about T2DM and subsequently finding ways to communicate that information to people affected by or at risk of the disease. One hundred and sixty-four questions were collected from 120 patients attending outpatient diabetes clinics and 300 questions from 100 members of the public through the Amazon Mechanical Turk crowdsourcing platform. Twenty-three general and diabetes-specific topics and five phases of disease progression were identified; these were used to manually categorize the questions. Analyses were performed to determine which topics, if any, were significant predictors of a question's being asked by a patient or the public, and similarly for questions from a woman or a man. Further analysis identified the individual topics that were assigned significantly more often to the crowdsourced or clinic questions. These were Causes (CI: [-0.07, -0.03], p < .001), Risk Factors ([-0.08, -0.03], p < .001), Prevention ([-0.06, -0.02], p < .001), Diagnosis ([-0.05, -0.02], p < .001), and Distribution of a Disease in a Population ([-0.05,-0.01], p = .0016) for the crowdsourced questions and Treatment ([0.03, 0.01], p = .0019), Disease Complications ([0.02, 0.07], p < .001), and Psychosocial ([0.05, 0.1], p < .001) for the clinic questions. No highly significant gender-specific topics emerged in our study, but questions about Weight were more likely to come from women and Psychosocial questions from men. There were significantly more crowdsourced questions about the time Prior to any Diagnosis ([(-0.11, -0.04], p = .0013) and significantly more clinic questions about Health Maintenance and Prevention after diagnosis ([0.07. 0.17], p < .001). A descriptive analysis pointed to the value provided by the specificity of questions, their potential to disclose emotions behind questions, and the as-yet unrecognized information needs they can reveal. Large-scale collection of questions from patients across the spectrum of T2DM progression and from the public-a significant percentage of whom are likely to be as yet undiagnosed-is expected to yield further valuable insights

    Correlation Of Terrestrial gamma flashes, Electric fields, and Lightning strikes (COTEL) in thunderstorms using networked balloon payloads developed by university and community college students

    High energy gamma ray flashes from terrestrial sources have been observed by satellites for decades, but the actual mechanism, assumed to be thunderstorm lightning, has yet to be fully characterized. The goal of COTEL, funded by NASA through the University Student Instrument Project (USIP) program, is to correlate in time TGF events, lightning strikes, and electric fields inside of thunderstorms. This will be accomplished using a small network of balloon-borne payloads suspended in and around thunderstorm environments. The payloads will detect and timestamp gamma radiation bursts, lightning strikes, and the intensity of localized electric fields. While in flight, data collected by the payloads will be transmitted to a ground station in real-time and will be analyzed post-flight to investigate potential correlations between lightning, TGFs, and electric fields. The ground station system that will be used for COTEL was developed for the Eclipse 2017 ballooning project, and was used during flight operations on the day of the eclipse. The COTEL student team is in its second year of effort having spent the first year developing the basic balloon payloads and ground tracking system. Currently the team is focusing on prototype electric field and gamma radiation detectors. Testing and development of these systems will continue into 2018, and flight operations will take place during the spring 2018 Louisiana thunderstorm season. The poster will cover the student team effort in developing said system, an overview of the system architecture, balloon flight tests conducted to date, preliminary results from prototype detectors, and future plans

    A qualitative study exploring midlife women’s stages of change from domestic violence towards freedom

    Gold OABackground Domestic Violence (DV) remains a significant global health problem for women in contemporary society. Existing literature on midlife women’s experiences of domestic violence is limited and focuses on health implications. Leaving a violent relationship is a dynamic process that often requires multiple attempts and separations prior to final termination. The aim of this study was to explore the process of leaving a violent relationship for midlife women. Methods This qualitative study involved fifteen women aged between 40–55 who had accessed residential and non-residential community support services for domestic violence within the UK. Community-based support agencies provided these women with access to letters of invitation and participant information sheet explaining the study. The women notified agency staff who contacted the research team to arrange a mutually convenient time to meet within a safe place for both the women and researchers. It was stressed to all potential participants that no identifiable information would be shared with the agency staff. Women were considered survivors of DV if they defined themselves as such. Data were gathered through semi structured interviews, transcribed verbatim and thematically analysed. Results Midlife women appear to differ from younger women by transitioning quickly though the stages of change, moving rapidly through the breaking free onto the maintenance stage. This rapid transition is the resultant effect of living with long-term violence causing a shift in the women’s perception towards the violent partner, with an associated reclamation of power from within the violent relationship. A realisation that rapid departure from the violence may be critical in terms of personal safety, and the realisation that there was something ‘wrong’ within the relationship, a ‘day of dawning’ that had not been apparent previously appears to positively affect the trajectory of leaving. Conclusions Midlife women appeared to navigate through the stages of change in a rapid linear process, forging ahead and exiting the relationship with certainty and without considering options. Whilst these findings appear to differ from younger women’s process of leaving, further research is needed to explore and understand the optimum time for intervention and support to maximise midlife women’s opportunities to escape an abusive partner, before being reflected appropriately in policy and practice.This study received funding from The Research and Knowledge Transfer Office, The University of Chester, and from the Western Australian Health Promotion Foundation – ‘Healthway
